Janus Kinase Inhibitor (JAK) can be referred to as medications that inhibit the abnormality of JAK families such as JAK1, JAK2, JAK3, and TYK2. These drugs have proven to be extremely helpful in the treatment of autoimmune diseases such as rheumatoid arthritis and certain cancer. Janus kinase is a family of intracellular, non-receptor tyrosine kinases that transduce cytokine-mediated signals via the JAK-STAT pathway. Janus kinase inhibitors works by inhibiting the activity of one or more of the JAK family, interfering with the JAK-STAT signaling pathway.

Janus kinase inhibitors play an important role in curing and preventing diseases such as cancer and rheumatoid arthritis. Thus, with the increasing prevalence of such diseases, the demand for Janus kinase inhibitors is also increasing with a rapid pace. According to the American Cancer Society, in 2021, there will be around 1.9 million new cancer cases diagnosed and over 608,570 cancer deaths in the United States. According to the Arthritis Foundation, around 1 in 3 people aged between 18 and 64 have arthritis.

Moreover, the number of adults in the U.S. with doctor-diagnosed arthritis is estimated to rise by 49% to reach 78.4 million (25.9% of all adults) by 2040. This in turn is expected to increase the demand for the Janus kinase inhibitors in this region. It is oral treatment that have become widely available and provide efficacious long-term oral biologic option in rheumatoid arthritis. Janus kinase inhibitors are the latest drug class of disease-modifying medication to emerge for the treatment of rheumatoid arthritis (RA).

JAK inhibitors belong to a family of medicine called disease-modifying antirheumatic drugs (DMARDs). Janus kinase inhibitors, such as upadacitinib (Rinvoq), tofacitinib (Xeljanz), and baricitinib (Olumiant), are approved by the U.S. Food and Drug Administration (FDA) to treat rheumatoid arthritis. Studies suggest JAK inhibitors are just as effective as biologic drugs.
